In Short: Two studies from Karolinska Institutet in Sweden reveal how genetic and environmental factors contribute to the debilitating condition.

Researchers analyzed genetic information from more than 1,500 people and examined blood, skin cells, and DNA from smaller groups of patients and healthy controls.
The studies, published in the journal Cephalalgia, strengthened the evidence for seven genes previously connected to cluster headache. Six of these genes either function in immune cells or participate in inflammatory signaling.
The investigation also showed differences in markers related to DNA methylation, a chemical process that helps regulate gene activity without changing the underlying DNA sequence.
In a separate study, researchers investigated whether exposure to substances in tobacco smoke affects the regulation of genes in people with cluster headaches. They found that inflammation, altered gene regulation, and increased exposure to toxic substances may all contribute to the development of cluster headaches.
